FDA Draft Guidance: Container Closure Systems for Human Drugs & Biological Products

The U.S. FDA has issued a Draft Guidance for Industry (August 2026) outlining a risk-based framework for evaluating Container Closure Systems (CCS) used for human drugs and biological products.

🔍 Key areas covered:
• Safety and compatibility of packaging materials
• Protection against moisture, light, gases, contamination and leakage
• Container Closure Integrity and functional performance
• Manufacturing-process impacts such as sterilization and depyrogenation
• Storage, handling and transportation considerations
• Extractables & Leachables assessment
• Quality assessment, qualification and quality control
• Stability testing and shipping/handling studies
• Considerations for different dosage forms and routes of administration
• CCS information for marketing and investigational applications and post-approval changes

The guidance emphasizes that a container closure system suitable for one drug product cannot automatically be considered suitable for another. CCS selection and qualification should be based on product-specific risks, formulation, materials, manufacturing processes and intended clinical use.

📌 Important: This is a Draft Guidance — Not for Implementation and represents FDA's current thinking for comment purposes.

A valuable reference for professionals working in Pharmaceutical Packaging, QA, QC, CMC, Regulatory Affairs, Product Development and Manufacturing.

Validating Medical Packaging

According to the FDA Quality System Regulations, manufacturers must ensure that "device packaging and shipping containers are designed and constructed to protect the device from alteration or damage during the customary conditions of processing, storage, handling, and distribution." As specific as this statement is, the FDA does not provide instructions on how to achieve their standards. Validating Medical Packaging demystifies the validation procedure for medical device packaging by providing specific examples and templates for creating and maintaining a validation file. About the author: Ronald Pilchik has over 30 years of experience in the healthcare manufacturing industry. As principal of the Techmark Group, he provided consulting services to diverse multinational manufacturers on selecting, developing, and validating medical packaging compatible with their sterilization requirements. He has been chairman of the healthcare packaging section of TAPPI, the packaging committee of HIMA, and a member of AAMI's ISO198 working group on packaging. Mr. Pilchik is program chairman of the HealthPack Conference Series on medical device packaging.

Standardizing Medication Labels

Medications are an important component of health care, but each year their misuse results in over a million adverse drug events that lead to office and emergency room visits as well as hospitalizations and, in some cases, death. As a patient's most tangible source of information about what drug has been prescribed and how that drug is to be taken, the label on a container of prescription medication is a crucial line of defense against such medication safety problems, yet almost half of all patients misunderstand label instructions about how to take their medicines. Standardizing Medication Labels: Confusing Patients Less is the summary of a workshop, held in Washington, D.C. on October 12, 2007, that was organized to examine what is known about how medication container labeling affects patient safety and to discuss approaches to addressing identified problems.
